Observación mediante microscopio electrónico de las fases de contaminación y esporulación de Plasmopara viticola

Resumen
Observación mediante microscopio electrónico de las fases de contaminación y esporulación de Plasmopara viticola, en un genotipo resistente (Vitis rupestris) y otro susceptible (Vitis vinifera L, cv. Albariño). Se inocularon hojas de Vitis vinifera L. cv. Albariño (susceptible) y V. rupestris (resistente) con Plasmopara viticola. A continuación, se observaron mediante microscopía electrónica de barrido a baja temperatura (LT–SEM), las fases de contaminación y esporulación del hongo. A las 24hpi (horas post inoculación), el genotipo resistente (V. rupestris), presentaba pocos estomas infectados y a veces se observaban, en algunos de ellos, secreciones que rodeaban a las zoosporas. La formación de esporangióforos era menor y los esporangios eran escasos y más pequeños. Por el contrario, en el genotipo susceptible (Albariño), a las 24hpi ya había un gran número de estomas infectados por entre 2 y 5 zoosporas. La vesícula subestomática era perfectamente visible y no se observaron secrecciones alrededor de las zoosporas. A las 72hpi había muchos esporangióforos y esporangios que ya estaban libres. En general el desarrollo del hongo en el genotipo susceptible fue muy superior s que en el genotipo resistente.

Abstract
Observation of the Plasmopara viticola contamination and sporulation stages by electron microscopy, on a resistant genotype and another susceptible. Leaves of Vitis vinifera L. cv. Albariño (susceptible) and V. rupestris (resistant) were inoculated with Plasmopara viticola. Both contamination and sporulation stages were then examined by low temperature scanning electron microscopy (LT–SEM). Only few infected stomata were observed, twenty–four hours after inoculation, on the resistant genotype (V. rupestris), showing in some cases secretions surrounding the zoospores. On the sensitive genotype (cv. Albariño), a high number of infected stomata were detected at the same time after inoculation, with 2 to 5 zoospores per infected stomata. The substomatic vesicle was clearly visible with no secretion around the zoospores. After seventy–two hours there were a lot of sporangiophores and sporangia spread on the leaf surface at the sensitive genotype, while the development of sporangiophores was weaker on the resistant genotype, with fewer and smaller sporangia.
